I found this review article to be very well written and informative. The most interesting comments I pulled out of it had to due with the relatively poor in vitro-in vivo correlations observed with Pgp studies involving digoxin. The report that in their analysis, the correlations used (and outlined in an FDA Guidance) generate quite a high percentage of false positives (58%) and negatives (41%). I would highly recommend this article to anyone working in Pharma.
